Multinational phase 3 trial of rucaparib or physician’s choice for mCRPC in patients with BRCA1, BRCA2, or ATM alteration
Prof. Karim Fizazi
The multinational phase 3 TRITON3 trial recruited patients with metastatic castration-resistant prostate cancer and a BRCA1, BRCA2, or ATM alteration, who had progression of disease after treatment with a second-generation androgen-receptor pathway inhibitor. Patients were randomized to rucaparib or physician’s choice control medication (docetaxel, abiraterone, or enzalutamide). Among the patients with a BRCA1 or BRCA2 alteration the imaging-based progression-free survival
was significantly longer with rucaparib than with physician’s choice. This was not seen among patients with an ATM alteration.
