Multinational phase 3 trial of pirtobrutinib for covalent BTK inhibitor-pretreated CLL/SLL

The multinational phase 3 BRUIN CLL-321 trial compared pirtobrutinib versus investigator’s choice (IC) of idelalisib-rituximab or bendamustine-rituximab among CLL/CLL patients who were previously treated with a covalent Bruton tyrosine kinase inhibitor. Pirtobrutinib versus IC was associated with improved progression-free survival, improved time to next treatment (24.0 months versus 10.9 months; HR 0.37; 95% CI 0.25-0.52), and favorable tolerability (treatment discontinuation due to AE 17.2% versus 34.9%).
