Multinational phase 3 trial of lenalidomide plus rituximab for R/R indolent hon-Hodgkin lymphoma: 5-year follow-up of AUGMENT

The multinational phase 3 AUGMENT trial demonstrated improved efficacy for lenalidomide plus rituximab (R2) versus rituximab plus placebo (R-placebo) in patients with relapsed or refractory indolent non-Hodgkin lymphoma (R/R iNHL). Long-term follow-up results from AUGMENT have now been published. A total of 358 patients with R/R grade 1 to 3a iNHL were randomized 1:1 to R2 or R-placebo. At long-term (median 65.9 months) follow-up, progression-free survival (HR 0.50; 95% CI 0.38-0.66) and overall survival (0.59; 0.37-0.95) were improved with R2 versus R-placebo. Improved PFS and OS with R2 versus R-placebo were also observed among the 295 patients with follicular lymphoma and patients aged 70 years or older. Safety findings were consistent with the primary analysis. The authors conclude that with follow-up of longer than 5 years, data from the AUGMENT trial continue to support the use of R2 as a standard of care for patients with R/R iNHL.
