Multinational phase 3 trial of ipatasertib plus abiraterone and prednisolone for mCRPC with or without PTEN-loss

The multinational phase 3 study IPATential150 randomized patients with mCRPC to ipatasertib-abiraterone-prednisolone or placebo-abiraterone-prednisolone. Tumor PTEN-loss was a stratification factor. The median rPFS was significantly longer with ipatasertib-abiraterone-prednisolone than with placebo-abiraterone-prednisolone among the patients with PTEN-loss tumors but not among all patients. The authors conclude that combined AKT and androgen-receptor signalling pathway inhibition with ipatasertib and abiraterone is a potential treatment for men with PTEN-loss mCRPC, a population with a poor prognosis.
