The phase III three-arm POSEIDON trial of patients with first-line metastatic non-small cell lung cancer (mNSCLC) evaluated the efficacy of tremelimumab plus durvalumab plus chemotherapy (T + D + CT) and durvalumab plus chemotherapy (D + CT) versus chemotherapy alone (CT). The trial showed that D + CT significantly improved progression-free survival versus CT, with a positive trend for improved overall survival that did not reach statistical significance. T + D + CT significantly improved progression-free survival and overall survival versus CT, without clinically meaningful increase in tolerability burden. The authors conclude that T + D + CT may represent a new first-line treatment option in mNSCLC.
