Multinational phase 3 trial of chemotherapy with or without amivantamab for EGFR-mutant aNSCLC after osimertinib: PROs from MARIPOSA-2

The multinational phase 3 MARIPOSA-2 trial demonstrated improved progression-free survival with amivantamab plus chemotherapy (n=131) compared with chemotherapy alone (n=263) in patients with EGFR-mutated locally advanced or metastatic non-small cell lung cancer (LA/M NSCLC) with disease progression on or after osimertinib. Pation-reported outcomes (PROs) and time to symptomatic progression (TTSP) have now been published. PRO scores were stable over time with little or no group differences in change from baseline. At 6 months post-baseline, 55.1% of patients in the amivantamab-chemotherapy group reported improved or stable QLQ-C30 global health status/ quality or life scores, compared with 29.1% in the chemotherapy group. TTSP was prolonged with amivantamab-chemotherapy versus chemotherapy alone (HR 0.73; p=0.0259). The authors conclude that the oncological benefits of adding amivantamab to chemotherapy were achieved without compromising health-related quality of life, and that adding amivantamab resulted in prolonged TTSP.
