The multinational phase 3 CONTACT-01 trial included patients with metastatic non-small cell lung cancer (mNSCLC) who developed disease progression after concurrent or sequential treatment with anti-PD-(L)1 and platinum-containing chemotherapy. Patients were randomized 1:1 to atezolizumab plus cabozantinib or docetaxel. Median overall survival (primary end point) was not significantly longer with atezolizumab plus cabozantinib than with docetaxel, although median progression-free survival was significantly longer with atezolizumab plus cabozantinib than with docetaxel. Grade 3 or 4 treatment-related adverse events occurred in 39.5% versus 34.7%, and grade 5 TRAEs in 2.2% versus 0.6%. The authors conclude that atezolizumab plus cabozantinib after disease progression following anti-PD-(L)1 and chemotherapy did not improve OS compared with docetaxel.
