Multinational phase 3 trial of addition of olaparib to first-line abiraterone for mCRPC: final overall survival results of PROpel

The multinational phase 3 PROpel trial, at 126 centers in 17 countries, evaluated addition of olaparib versus placebo to first-line abiraterone plus prednisone or prednisolone for metastatic castration-resistant prostate cancer (mCRPC). The study met its primary end point showing statistically significant improvement in radiographic progression-free survival in the olaparib group. Final overall survival analysis of the trial found that addition of olaparib to abiraterone did not significantly improve overall survival. Addition of olaparib was associated with improved subsequent anti-cancer treatment-free survival and improved second progression-free survival. Addition of olaparib was associated with longer overall survival among the HRR-mutated patients and the BRCA-mutated patients.
