The multinational phase 3 KRYSTAL-12 trial compared adagrasib versus docetaxel in patients with advanced NSLSC with KRAS-G12C mutation who previously received chemotherapy and immunotherapy. In the ITT population (median follow-up 7.2 months), the median progression-free survival was 5.5 months with adagrasib and 3.8 months with docetaxel (HR 0.58; p<0.0001). Grade 3 or higher treatment-related adverse events occurred in 47% versus 46%; grade 5 TRAEs in 4 of 301 patients versus 1 of 140 patients. The authors conclude that adagrasib demonstrated a statistically significant improvement in progression-free survival over docetaxel in patients with previously treated KRAS-G12C-mutated aNSCLC, without new safety signals.
