Multinational phase 3 trial of abiraterone plus olaparib for metastatic castration-resistant prostate cancer
Prof. Noel Clarke
Preclinical studies and results of a phase 2 trial of abiraterone and olaparib suggest a combined antitumor effect of both agents for metastatic castration-resistant prostate cancer (mCRPC). The multinational phase 3 PROpel trial compared first-line abiraterone plus olaparib versus abiraterone plus placebo for mCRPC. The primary end point was imaging-based progression-free survival (ibPFS). The ibPFS (A: investigator assessed; B: blinded independent central review) was significantly longer with abiraterone plus olaparib than with abiraterone plus placebo. Overall survival data were premature. The safety profile of abiraterone plus olaparib was consistent with the known safety profiles of the individual agents.
