Multinational phase 3 study of pola-R-CHP versus R-CHOP for newly diagnosed DLBCL
Prof. Hervé Tilly
The current standard of care for newly diagnosed diffuse large B-cell lymphoma (ND DLBCL) is R-CHOP; however, approximately 40% of patients are not cured. Polatuzumab vedoting is a CD79b-targeting antibody-drug conjugate. The multinational phase 3 POLARIX study compared polatuzumab vedotin, rituximab, cyclophosphamide, doxorubicin, and prednisone (pola-R-CHP) versus R-CHOP for newly diagnosed DLBCL. The pola-R-CHP combination demonstrated a 27% reduction in relative risk of disease progression, relapse, or death compared with R-CHOP, with a similar safety profile.
