Leptomeningeal metastatic disease (LMD) is a severe complication of solid cancers. The antibody-drug conjugate patritumab deruxtecan (HER3-DXd) targets HER3 which is involved in CNS metastasis. Cohort 3 of the multinational phase 2 TUXEDO-3 trial evaluated HER3-DXd for LMD of any solid tumor. The primary endpoint was met with 65% of patients alive after 3 months. Neurological symptoms and quality of life remained stable or improved during study treatment. The authors conclude that TUXEDO-3 showed clinically relevant activity of HER3-DXd in patients with LMD.
