Multinational phase 2 trial of first-line encorafenib plus binimetinib and cetuximab for metastatic colorectal cancer with BRAF V600E mutation
Prof. Eric van Cutsem
The multinational phase 2 ANCHOR CRC trial evaluated the combination of encorafenib (BRAF inhibitor), binimetinib (MAPK inhibitor), and cetuximab (anti-EGFR monoclonal antibody) as first-line treatment for patients with metastatic colorectal cancer (mCRC) with BRAF V600E mutation. The trial met its primary end point with a confirmed objective response rate of 47.4% (95% CI 37.0-57.9).The median progression-free survival was 5.8 months, and the median overall survival was 18.3 months. The treatment was well tolerated, with no unexpected toxicities. The authors conclude that the combination may be an option for patients not eligible to receive standard first-line therapy.
