FGFR alterations are reported across various malignancies and might act as oncogenic drivers in multiple histologies. Erdafitinib is an oral, selective pan-FGFR tyrosine kinase inhibitor with activity in FGFR-altered advanced urothelial carcinoma. The multinational phase 2 RAGNAR trial (156 centers in 15 countries) evaluated erdafitinib for advanced solid tumors of any histology (except urothelial carcinoma) with FGFR1-4 alterations. In the primary cohort of RAGNAR, objective response was seen in 30% of patients (95% CI 24-36) across 16 distinct tumor types. Toxicity was acceptable. The authors conclude that these results support the continued development of FGFR inhibitors for patients with advanced solid tumors.
