Multinational phase 2 trial of dabrafenib plus trametinib for relapsed/refractory BRAF V600-mutant pediatric high-grade glioma
Prof. Darren Hargrave
BRAF V600 mutation is detected in 5%-10% of pediatric high-grade gliomas (pHGGs), and effective treatments are limited. A multinational phase 2 trial evaluated the combination of dabrafenib and trametinib for relapsed or refractory BRAF V600-mutant pHGG. The combination improved the overall response rate versus previous trials of chemotherapy for molecularly unselected patient with pHGG, and was associated with durable responses and encouraging progression-free survival. The authors conclude that dabrafenib plus trametinib is a promising targeted therapy option for patients with relapsed/refractory BRAF V600-mutant pHGG.
