The multinational phase 2 TALAPRO-1 study evaluated the PARP inhibitor talazoparib for heavily pretreated metastatic castration-resistant prostate cancer with DNA damage response alterations in genes involved in homologous recombination repair. Objective response was seen in 29.8% of the evaluable patients, while serious treatment-emergent adverse events were reported in 34% of patients. The authors conclude that talazoparib showed durable antitumor efficacy, with a favorable benefit-risk profile.
