Multinational phase 2 study of mirvetuximab soravtansine for platinum-resistant ovarian cancer with high expression of folate receptor alpha

Single-agent chemotherapies have limited activity and considerable toxicity in patients with platinum-resistant epithelial ovarian cancer (PROC). Mirvetuximab soravtansine (MIRV) is an antibody-drug conjugate targeting folate receptor α (FRα). The multinational phase 2 SORAYA study evaluated MIRV for PROC with high FRα expression after one to three prior therapies, including bevacizumab. Response to MIRV was seen in 32.4% of patients (95% CI 23.6-42.2) with complete response in 4.8%. The median duration of response was 6.9 months (95% CI 5.6-9.7). Treatment-related adverse events led to dose delays in 33% of patients, to dose reductions in 20%, and to discontinuation in 9%. The authors conclude that MIRV had clinically meaningful antitumor activity and safety in patients with FRα-high PROC who had received up to three prior therapies.
