The multinational phase 2 PCYC-1123-CA study evaluated the ibrutinib-lenalidomide-rituximab (the iR2 regimen) for relapsed or refractory non-GCB DLBCL in patients ineligible for stem cell transplantation. The ORR was 49% with complete response in 28% of the patients. Grade 3-4 AEs were reported for 91% of patients, and grade 5 AEs for 13%. The authors conclude that the most frequent AEs were consistent with known safety profiles of the individual drugs, and that the iR2 regimen demonstrated durable antitumor activity.
