Multinational phase 2 study of epcoritamab for relapsed or refractory follicular lymphoma

Epcoritamab is a novel CD3 x CD20 bispecific antibody. A phase 2 cohort of the multinational, multicohort EPCORE NHL-1 trial evaluated subcutaneous epcoritamab as third or later line therapy for relapsed or refractory follicular lymphoma (R/R FL). The overall response rate was 82.0% (95% CI 74.3-88.3) with complete response in 62.5% (53.5-70.9). The most common grade 3 or 4 treatment-emergent adverse event was neutropenia (25% of patients). Grade 1 or 2 cytokine release syndrome was reported in 65% and grade 3 CRS in 2%. Grade 1 immune effector cell-associated neurotoxicity syndrome was reported in 4% and grade 2 ICANS in 2%. The authors conclude that epcoritamab monotherapy had clinically meaningful activity and manageable safety among patients with R/R FL.
