Enfortumab vedotin )EV) is a nectin-4-directed antibody-drug conjugate with established efficacy in urothelial carcinoma. The multinational phase 2 EV-202 study evaluated EV in adult patients with previously treated locally advanced or metastatic HR-positive/HER2-negative breast cancer or triple-negative breast cancer who had not received more than 2 previous lines of cytotoxic therapy in the LA/M setting. Confirmed objective response rate and disease control rate were 15.6% and 51.1%, respectively, in the HR+/HER2- cohort (n=45) and 19.0% and 57.1%, respectively, in the TNBC cohort (n=42). Median progression-free survival and overall survival were 5.39 months and 19.75 months, respectively, in the HR+/HER2- cohort and 3.52 months and 12.91 months, respectively, in the TNBC cohort. Safety profile was consistent with previous reports. These results suggest EV could be a new treatment possibility for certain patients with advanced breast cancer, and more research is warranted.
