Targeted therapy and immunotherapy have shown intracranial activity in melanoma with CNS metastases, but there remains an unmet need, particularly for patients with symptomatic CNS metastases. The phase 2 TRICOTEL study (65 patients of 21 centers in 7 countries) found that the combination of atezolizumab, vemurafenib, and cobimetinib resulted in intracranial objective response in 42% (95% CI 29-54) of patients with BRAFV600-mutated melanoma. The combination had promising intracranial and extracranial activity in this patient cohort.
