Multinational phase 2-3 study of entrectenib in ROS1- positive advanced non-small cell lung cancer

Comprehensive biomarker testing is recommended for patients with advanced non-small cell lung cancer (aNSCLC), but tissue availability can limit testing. The multinational phase 2-3 Blood First Assay Screening Trial (BFAST) evaluates the efficacy and safety of multiple therapies in patients with aNSCLC and targetable alterations targeted by liquid biopsy. Cohort D of the trial recruted adult patients with aNSCLC with ROS1-positivity detected in liquid biopsies, who received entrectinib 600 mg daily. The confirmed objective response rated by investigator was 81.5%, meeting the primary endpoint. The figure shows results for secondary endpoints. The safety profile of entrectinib was consistent with previous reports. The authors conclude that the integration of liquid biopsies into clinical practice provides patients with a less invasive diagnostic method than tissue-based testing and has faster turnaround times that may expedite the reaching of clinical decisions in the aNSCLC setting.
