Multinational phase 1b study of telisotuzumab vedotin plus erlotinib for c-Met protein-expressing NSCLC

Overexpression of c-Met protein and epidermal growth factor receptor (EGFR) mutations can co-occur in non-small cell lung cancer (NSCLC), providing strong rationale for dual targeting. Telisotuzumab vedotin (Teliso-V) is a first-in-class antibody-drug conjugate targeting c-Met. A multinational phase 1b study evaluated the combination of Teliso-V and erlotinib for c-Met-positive NSCLC pretreated with EFGR TKI. Neuropathies were the most common any-grade adverse events (57%). Median PFS was 5.9 months. Among patients with EGFR-activating mutation the ORR was 32.1%, and among patients with EGFR-activating mutation who were c-Met high the ORR was 52.6%. The authors conclude that Teliso-V plus erlotinib showwed encouraging antitumor activity and acceptable toxicity for EGFR TKI-pretreated NSCLC with EGFR-activating mutation and c-Met overexpression.
