A multinational phase 1b-2 trial evaluated the combination of glofitamab and polatuzumab vedotin (Glofit-Pola) in a population with refractory or relapsed large B-cell lymphoma (R/R LBCL). Patients had received a median of 2 (range 1-7) previous lines of treatment (previous CAR T-cell therapy: 21.7%). The independent review committee-assessed overall response rate was 78.3% (complete response rate 59.7%). Median progression-free survival and overall survival were 12.3 and 33.8 months, respectively. The most common adverse event was cytokine release syndrome, and the events were mainly low-grade. The authors conclude that Glofit-Pola demonstrated high efficacy and durable responses, with manageable safety, in heavily pretreated patients with R/R LBCL.
