Patritumab deruxtecan (HER3-DXd) is a HER3-directed antibody-drug conjugate that has shown durable antitumor activity in previously treated patients with EGFR-mutated advanced non-small cell lung cancer (aNSCLC). The multinational phase 1 U31402-AU102 trial evaluated HER3-DXd for previously treated aNSCLC with other or no identified driver genomic alterations. The confirmed objective response rate was 27.7% (95% CI 15.6-42.6) and the median duration of response was 8.1 monthts (4.2-NE). The median progression-free survival and overall survival were 5.5 months (95% CI 4.0-11.2) and 15.2 months (10.8-17.7), respectively. The rate of study drug discontinuation associated with treatment-emergent adverse events was 12.8%. The authors conclude that the previously reported efficacy and safety of HER3-DXd in pretreated patients with EGFR-mutated aNSCLC were also observed in those with other NSCLC subtypes.
