Praluzatamab ravtansine consists of an anti-CD166 monoclonal antibody conjugated to DM4, with a protease-cleavable linker that can be cleaved in the microenvironment of CD166 expressing tumors. A multinational phase 1-2 study evaluated praluzatamab ravtansine for advanced solid tumors after at least 2 previous lines of therapy. The treatment resulted in regressions in a variety of tumor types. Among patients with HR-positive/HER2-non-amplified breast cancer 9% had partial response and 45% had stable disease. The authors conclude that CD166 is a novel, ubiquitously expressed target. Praluzatamab ravtansine had both translational and clinical activity.
