Multinational phase 1-2 study of lenvatinib with etoposide plus ifosfamide for refractory or relapsed osteosarcoma

Tyrosine kinase inhibitors have show activity in osteosarcoma and might enhance the efficacy of chemotherapy. The multinational phase 1-2 ITCC-050 study evaluated the TKI lenvatinib combined with etoposide plus ifosfamide chemotherapy for refractory or relapsed osteosarcoma. The combination resulted in 4 months progression-free survival in 18 of 35 patients (51%; 95% CI 34-69) treated at the recommended phase 2 dose. There were no new safety signals. The authors conclude that these findings warrant further investigation.
