Multicenter study of neoadjuvant niraparib for HER2-negative, BRCA-mutated, resectable breast cancer
Dr. Steven Isakoff
Niraparib is a PARP-1/2 inhibitor approved for advanced endometrial cancer. A multicenter pilot study in the USA evaluated neoadjuvant niraparib for HER2-negative, BRCA-mutated, resectable breast cancer. After 2 cycles, tumor response (≥30% reduction from baseline) by MRI was seen in 90.5% of patients. pCR after 2-6 cycles was seen in 40% of patients receiving only niraparib. High niraparib intratumoral concentration was observed. No new safety signals were identified.
