The multicenter CARTBCMA-HCB-01 trial in Spain evaluated fractionated initial infusion and booster dose of ARI0002h, a humanised, BCMA-directed CAR T-cell therapy, for patients with relapsed or refractory multiple myeloma. The ORR 100 days after the first infusion was 100%, including 50% of patients with complete response. Cytokine release syndrome was seen in 80%; all cases were grade 1 or 2. There were no cases of neurotoxic events. The authors conclude that ARI0002h administered in a fractionated manner followed by a booster dose after three months can provide deep and sustained responses in patients with RRMM, with low toxicity, especially in terms of neurologic events.
