Multicenter retrospective study of characteristics of patients with NRAS-mutated non-small cell lung cancer

NRAS mutations have been observed in less than 1% of non-small cell lung cancers. Clinical data regarding this rare subset are scarce, and response to treatment has never been reported. A retrospective study in 14 center in France included 164 consecutive patients with NRAS mutated NSCLC. Most patients were male (64.6%), current or former smokers (91.5%), and had stage IV disease at diagnosis (81.7%). Most mutations were seen in NRAS codon 61. Median overall survival of stage IV patients was 15.3 months (95% CI 9.9-27.6). Among stage IV patients, objective response rate and median progression-free survival were 45% respectively 5.8 months with doublet chemotherapy, 35% respectively 6.9 months with immune checkpoint inhibition, and 70% respectively 8.6 months with a combination of chemotherapy and ICI.
