CAR T-cell therapy has reshaped the treatment paradigm for R/R DLBCL, but its role in R/R CNSL is not clear. A retrospective study ar four centers in the USA and Israel evaluated axi-cel, tisa-cel, liso-cel or point of care (POC) CD19-CAR T-cell products for R/R CNSL. At 100 days post infusion, 65% of patients attained complete response. The median PFS was 7.5 months and the median OS was 19 months. Patients receiving tisa-cel had poorer outcomes than patients receiving axi-cel, and patients receiving liso-cel or POC product had better outcomes than patients receiving axi-cel. Toxicity profiles were consistent with what was seen in pivotal trials in R/R DLBCL. The authors conclude that these results demonstrate the feasibility and safety of anti-CD19 CAR-T therapy in CNSL.
