Multicenter randomized phase 2 trial of sacituzumab tirumotecan versus docetaxel for pretreated EGFR-mutated advanced non-small cell lung cancer

The multicenter randomized phase 2 OptiTROP-Lung03 trial in China compared the trophoblast cell surface antigen 2 directed antibody-drug conjugate sacituzumab tirumotecan (sac-TMT) versus docetaxel in patients with EGFR-mutated advanced NSCLC after failure of EGFR-TKI and platinum-based chemotherapy. Sac-TMT improved the objective response rate compared with docetaxel and also improved progression-free survival and overall survival. Grade ≥3 treatment-related adverse events were less frequent with sac-TMT than with docetaxel (56% versus 72%) with no new safety signals identified.
