Patients with no evidence of disease after metastasectomy for metastatic renal cell carcinoma (mRCC) are at high risk of recurrence. The multicenter phase 3 ECOG-ACRIN E2810 trial randomized 129 of such patients to pazopanib or placebo for 52 weeks. The 3-year disease-free survival rate was 27.4% in the pazopanib group and 21.9% in the placebo group (HR 0.90; p=0.29). The 3-year overall survival rate was 81.9% in the pazopanib group and 91.4% in the placebo group (HR 2.55; p=0,012). Health-related quality of life measures deteriorated in the pazopanib group during the treatment period. The authors conclude that among metastatic renal cell carcinoma patients who had no evidence of disease after metastasectomy, pazopanib compared with placebo was not associated with improved disease-free survival, and was associated with worse overall survival.
