Multicenter phase 3 trial of neoadjuvant camrelizumab versus placebo in combination with chemotherapy for early or locally advanced TNBC

Preferred neoadjuvant strategies for early or locally advanced triple-negative breast cancer (TNBC) include a 4-drug chemotherapy regimen containing anthracyclines, cyclophosphamide, taxanes, and platinum. Camrelizumab (anti-PD-1) has shown antitumor activity in advanced TNBC. The phase 3 CamRelief trial, at 40 centers in China, randomized patients with early or locally advanced TNBC to neoadjuvant camrelizumab plus chemotherapy versus placebo plus chemotherapy. In this trial that included 441 patients, camrelizumab plus chemotherapy, compared with placebo plus chemotherapy, significantly improve pathological complete response rate (56.8% versus 44.7%). The authors conclude that addition of camrelizumab to neoadjuvant chemotherapy for early or locally advanced TNBC resulted in improved pathological complete response rate (visual abstract).
