Glecirasib (JAB-21822) is a novel oral covalent KRAS-G12C inhibitor. A multicenter phase 2 trial in China evaluated glecirasib administered at 800 mg daily in patients with locally advanced or metastatic non-small cell lung cancer (NSCLC) with KRAS-G12C mutation. The primary endpoint was objective response rate assessed by an independent review committee. The ORR assessed by IRC was 47.9% (95% CI 38.5-57.3). The incidence of treatment-related adverse events of any grade was 97.5%; of grade 3 or 4 38.7%; and resulting in discontinuation 5%. There were no grade 5 TRAEs. The authors conclude that glecirasib exhibited promising efficacy and manageable safety among patients with locally advanced or metastatic NSCLC with KRAS-G12C mutation.
