Multicenter phase 2b study of MDNA55 for recurrent glioblastoma
Prof. John Sampson
The interleukine-4 receptor (IL4R) is overexpressed in glioblastoma and cells of the tumor microenvironment. High expression of IL4R is associated with poor clinical outcome. MDNA55 is an IL4R-targeting toxin. The multicenter phase 2b study MDNA55-05 evaluated MDNA55 for recurrent glioblastoma. Single dose intratumoral administration had an acceptable safety profile at doses up to 240 μg. Among patients treated with MDNA dose > 180 μg, tumor control was seen in 81%, with median overall survival 15 months and one-year OS of 55%. The authors conclude that MDNA55 demonstrated tumor control and promising survival.
