Multicenter phase 2b study of cediranib plus olaparib for recurrent platinum-resistant ovarian cancer without a germline BRCA1/2 mutation
Dr. Joyce Liu
Cytoreductive surgery and chemotherapy are the mainstay of first-line therapy for ovarian cancer (OC), but approximately 80% of women with advanced OC experience a recurrence within 3 years of diagnosis, and almost all patients with recurrent disease ultimately develop platinum resistance. The multicenter phase 2b CONCERTO trial, in 25 US centers, evaluated the combination of the VEGF TKI cediranib and the PARP inhibitor olaparib for heavily pretreated recurrent platinum-resistant ovarian cancer in patients without a germline BRCA
mutation. The treatment resulted in ORR 15.3%, median PFS 5.1 months, and median OS 13.2 months.
