Multicenter phase 2a trial of SurVaxM and adjuvant temozolomide for newly diagnosed glioblastoma
Dr. Robert Fenstermaker
Despite intensive treatment, mortality among patients with newly diagnosed glioblastoma (nGBM) remains high. SurVaxM is a peptide vaccine targeting the cell-survival protein survivin, which is highly expressed by glioma cells. A multicenter phase 2a trial evaluated safety and efficacy of SurVaxM plus adjuvant temozolomide (TMZ) after surgery and chemoradiation for nGBM. The combination was well tolerated with no serious adverse event. Progression-free survival rate six months after diagnosis was 95.2%. Median PFS was 11.4 months and median OS was 25.9 months measured from first dose of SurVaxM. Patients generated survivin-specific immune responses, and antibody response was positively associated with survival.
