Multicenter phase 2 trial of upfront panitumumab plus mFOLFOX or panitumumab plus fluorouracil-leucovorin for RAS/BRAF-wt mCRC in elderly patients

There is an unmet need for first-line options for unresectable RAS/BRAF wild-type metastatic colorectal cancer in elderly patients (70 years or older). The multicenter phase 2 PANDA trial in Italy evaluated first-line panitumumab plus mFOLFOX (cohort A) or panitumumab plus fluorouracil and leucovorin (cohort B) for these patients. In boths cohorts patients received twelve cycles of the combination followed by panitumumab maintenance. The median progression-free survival was 9.6 months in cohort A and 9.0 months in cohort B. Rates of grade 2 or greater chemotherapy-related adverse events were 60% in cohort A and 37% in cohort B. The authors conclude that both combinations are reasonable first-line options for RAS/BRAF wild-type mCRC in elderly patients.
