Multicenter phase 2 trial of the WEE1 inhibitor adavosertib in refractory solid tumors harboring CCNE1 amplification
Prof. Siqing Fu
CCNE1
amplification is frequently seen in many types of malignancies, associated with resistance to chemotherapy and short survival. A multicenter phase 2 trial in the United States assessed the antitumor activity of the WEE1 kinase inhibitor adavosertib in patients with CCNE1-amplified, advanced refractory solid tumors. Adavosertib had a manageable toxicity profile and promising clinical activity (ORR 27%; median OS 9.9 months), especially in epithelial ovarian cancer (ORR 36%; median OS 14.9 months).
