Multicenter phase 2 trial of sacituzumab govitecan for metastatic solid tumors: results of the endometrial cancer cohort

Sacituzumab govitecan (SG) is a Trop-2-directed antibody-drug conjugate. The multicenter multicohort phase 2 TROPiCS-03 trial in the USA evaluates SG in patients with metastatic solid tumors. Results of the endometrial cancer cohorts have now been published. The cohort included patients who received a median of three (range 1-6) previous lines of therapy. The objective response rate was 22% (95% CI 11-38) with a median duration of response of 8.8 months (2.8-NE) while the clinical benefit rate was 32% (18-48). The median progression-free survival was 4.8 months (95% CI 2.8-9.8). Grade 3 or 4 treatment-related adverse events occurred in 73% of patients and led to discontinuation in 5%. Two of 41 patients died, deemed unrelated to SG. The authors conclude that SG had encouraging efficacy and manageable toxicity among heavily prertreated patients with advanced endometrial cancer.
