Multicenter phase 2 trial of pertuzumab plus trastuzumab for advanced biliary tract cancer with ERBB2/3 alterations

Targeted Agent and Profiling Utilization Registry (TAPUR) is a phase 2 basket trial evaluating the antitumor activity of commercially available targeted agents in patients with advanced cancer and genomic alterations known to be drug targets. One of the TAPUR cohorts consists of patients with advanced biliary tract cancer (aBTC) with ERBB2/3 amplification, overexpression, or mutation who were treated with the combination of pertuzumab and trastuzumab. Among 28 evaluable patients, one complete response and eight partial responses and two patients with stable disease for at least 16 weeks were seen, for a disease control rate of 40% (90% CI 27-100) and an overall response rate of 32% (95% CI 16-52). Four patients had at least one grade 3 adverse event or serious AE at least possibly related to treatment. The authors conclude that the combination of pertuzumab and trastuzumab had antitumor activity among patients with aBTC with ERBB2/3 alterations.
