The Targeted Agent and Profiling Utilization Registry (TAPUR) is a phase II basket trial evaluating the antitumor activity of commercially available targeted agents in patients with advanced cancer and genomic alterations known to be drug targets. One of the TAPUR cohorts consists of patients with advanced pancreatic cancer and BRCA1/2 mutations (germline or somatic). The median number of reported previous therapies was 3 (range 1-10). Olaparib twice daily (600 mg in tablets or 800 mg in capsules) resulted in disease control rate of 31% (90% CI 18-40) and objective response rate of 18% (95% CI 6-37). The median progression-free survival was 8 weeks (95% CI 8-15) and the median overall survival was 38 weeks (21-65). Ten percent of patients had at least one drug-related grade 3 adverse event or serious adverse event of anemia, fever, or oral mucositis. The authors conclude that olaparib showed antitumor activity in patients with advanced pancreatic cancer with BRCA1/2 mutations.
