The multicenter phase 2 OBI-1 trial in Poland evalutated obinutuzumab monotherapy as induction and maintenance in patients with relapsed or refractory Waldenström macroglobulinemia (RRWM). The best overall response rate was 65.2%, while the ORR was 51% at the end of induction and 65% after two years of maintenance treatment. Percentages of patients with very good partial response and complete remission increased during the maintenance phase. Median progression-free survival and overall survival were not reached, with 42-months PFS rate 65% and 66-month OS rate 74%. Toxicity was manageable. The authors conclude that obinutuzumab was active as induction and maintenance therapy for RRWM.
