A multicenter phase 2 trial in South Korea evaluated the third-generation EGFR-TKI lazertinib in patients with advanced non-small cell lung cancer harboring oncommon EGFR mutations. The objective response rate was 50% (95% CI 34.5-65.5), meeting the primary endpoint. The disease control rate was 88.9% (95% CI 74.1-96.2), the median duration of response was 15.1 months, and the median progression-free survival was 10.8 months. G719X mutation showed the highest response (ORR 61%, median PFS 20.3 months). Grade ≥ 3 treatment-emergent adverse events occurred in 33.3%. The authors conclude that lazertinib demonstrated promising efficacy and a manageable safety profile in patients with aNSCLC harboring uncommon EGFR mutations.
