Multicenter phase 2 trial of ixazomib, daratumumab, and low-dose dexamethasone for newly diagnosed multiple myeloma in intermediate-fit patients
Prof. Sonja Zweegman
The multicenter phase 2 HOVON-143 trial evaluated the combination of ixazomib, daratumumab, and low-dose dexamethasone for newly diagnosed multiple myeloma in intermediate-fit patients. The combination was safe and improved global quality of life. The ORR at end of induction was 71%. At median follow-up of 41.0 months, the median progression-free survival (panel A) and 3-year overall survival (panel B)
were 18.2 months and 83%, respectively. Efficacy was limited by ixazomib-induced toxicity, hampering long-term tolerability of this 3-drug regimen. The authors conclude that there is a need for more efficacious and tolerable regimens to improve outcomes of vulnerable intermediate-fit patients.
