Multicenter phase 2 trial of impact of lean body mass-based adjuvant oxaliplatin dose calculation on neurotoxicity in stage III colon cancer

Oxaliplatin-based chemotherapy often induces chronic peripheral neurotoxicity, significantly affecting clinical outcomes and quality of life. Oxaliplatin dose in usually based on the patient’s body surface (BSA); however, it is not clear whether dose adjustments based on lean body mass (LBM) could reduce oxaliplatin-induced peripheral neurotoxicity (OIPN) in patients with colon cancer. The multicenter phase 2 LEANOX trialin France randomized patients with resected stage 3 colon cancer to BSA-based oxaliplatin dose (arm 2) or LBM-based oxaliplatin dose (arm 3). The grade 2 or worse OPIN-free survival was significantly improved in arm 3 compared with arm 2, without compromising relapse-fre survival and overall survival.
