Multicenter phase 2 trial of hepatic arterial infusion pump chemotherapy for unresectable intrahepatic cholangiocarcinoma

The 3-year overall survival rate for advanced intrahepatic cholangiocarcinoma (iCCA) confined to the liver after systemic chemotherapy with gemcitabine-cisplatin (gem-cis) is only 3%. Hepatic arterial infusion pump (HAIP) chemotherapy with floxuridine aims to control the liver disease. The multicenter PUMP-2 trial in The Netherlands evaluated the combination of HAIP chemotherapy and systemic gem-cis for unresectable iCCA confined to the liver. The median OS was 22.3 months, and the 3-year OS rate was 31.5%. The 1-year OS rate of 80.0% was superior to the historical control of 47% with gem-cis alone (p<0.001).
