Multicenter phase 2 trial of glofitamab in refractory or relapsed diffuse large B cell lymphoma after failure of CAR-T cell therapy

A multicenter phase 2 trial in France evaluated th CD20-CD30 T cell-engaging bispecif antibody glofitamab in patients with diffuse large B cell lymphoma (DLBCL) after failure of CAR-T cell therapy. A total of 46 patients received at least on glofitamab infusion following obinutuzumab (anti-CD20 monoclonal antibody) pretreatment. After a median follow-up of 15,3 months, the median overall survival was 14.7 months (90% CI, 8.8-NR), meeting the primary endpoint. The median progression-free survival was 3.8 months (95% CI, 2.4-19.6), the best overall metabolic response rate was 76.1% and the best complete metabolic response rate was 45.7%. There was no grade 3 or higher cytokine release syndrome or neurotoxicity. The authors conclude that glofitamab improved OS in patients with R/R DLBCL after failure of CAR-T cell therapy, with a favorable safety profile.
