Multicenter phase 2 trial of garsorasib for KRAS G12C-mutated non-small cell lung cancer

Garsorasib is a potent KRASG12C inhibitor. A phase 2 trial at 43 centers in China evaluated garsorasib for KRASG12C-mutated non-small cell lung cancer previously treated with platinum-based chemotherapy and immune checkpoint inhibitors. The primary endpoint was objective response rate as assessed by an independent review committee (IRC-ORR). The median duration of follow-up was 7.9 months. The IRC-ORR was 50% (95% CI 41-59). Treatment-related adverse events were reported for 95% of patients; grade 3 or higher TRAEs for 50%. Most of the adverse events were well managed. The authors conclude that garsorasib was active with an acceptable and manageable safety profile among patients with previously treated KRASG12C-mutated non-small cell lung cancer.
